EFFECTIVENESS RATE OF THE TECHNIQUE FOR PRESERVING BACTERIA AND YEAST AT MINUS 20ºC

The preservation of microorganisms, such as bacteria and fungi, has a great importance for laboratories that work with these living beings on a daily basis, whether for teaching purposes, clinical or epidemiological research. Thus, this study aimed to evaluate the efficiency of the preservation and maintenance method adopted by the Laboratory of Practices of the Biomedical Clinic of Claretiano Faculdade de Boa Vista-Roraima. The culture collection of pathogenic microorganisms at Claretiano Faculdade has 30 microorganisms preserved at -20ºC in a common freezer. Procedures were carried out for the recovery of the preserved isolates from 2018 to 2023. To study the efficiency and recovery rate of the isolates, they were unfreezed and subjected to cultivation. The preserved isolates belong to groups of bacteria (gram positive and gram negative) and yeast fungi. The preservation of filamentous fungi will be done later. They are: Acinetobacter baumannii, Citrobacter freundii, Enterobacter spp, Enterococcus spp, Escherichia coli; Klebsiella pneumoniae (ESBL), Klebsiella pneumoniae (KPC), Morganella morganii, Proteus mirabilis, Pseudomonas aeruginosa NCTC, Pseudomonas aeruginosa clínica, Salmonella spp, Serratia marcescens, Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus agalactiae, Streptococcus pneumoniae, Streptococcus pyogenes, Candida albicans, Candida krusei, Candida tropicalis and 9 different strains of Cryptococcus spp. The methods evaluated were preservation by freezing at -20ºC for the 30 isolates. Our study showed efficiency and a 93% success rate. In a total of 28 isolates recovered with positive growth and preserved biochemical characteristics. Only 3 isolates did not show biochemical characteristics compatible with the initial identification. From this study, we concluded that this is a cheap technique and it presents significant results for the maintenance of these microorganisms for medium periods of time.
